Malvern OMNISEC multi-detection to monitor stability and aggregate formation

Size-exclusion chromatography (SEC) is an essential tool for the characterisation of biomolecules such as proteins for academic research or in the development of biopharmaceutical drugs. While single-detector SEC systems are widely used in characterisation, particularly for the detection of protein aggregates, multi-detector systems can increase the value of this technique by generating more accurate information, such as absolute molecular weight, or providing additional information on structure, oligomeric state and composition.

With additional detectors comes a wealth of additional information. Light scattering detectors, whether SEC-MALS (multi-angle light scattering), RALS (right-angle light scattering) or LALS (low-angle light scattering), measure absolute molecular weight with good accuracy. This allows oligomers and larger aggregates to be easily differentiated and quantified. Conjugate analyses allow the characterisation of PEGylated proteins, antibody-drug-conjugates (ADCs) and membrane proteins. A viscometer is used to study large changes in protein conformation.

While single detector systems offer high repeatability, differences between samples may not be visible when using UV-SEC alone. Using the Malvern Panalytical OMNISEC multi-detection SEC system, the addition of a light scattering detector enables absolute molecular weight measurements. This results in more complete understanding of protein and the types of aggregates formed, which enables better control over behaviour and stability. Better characterisation of proteins and better control of their production should result in a better product, meaning fewer quality issues, less waste and better control of manufacturing costs.
