Scientifica HoloStim-3D high-resolution multiphoton imager
Scientifica’s HoloStim-3D was designed to help perform more complex photostimulation experiments than are possible with random-access two-photon or widefield photostimulation. Users can discover the roles of neuronal subtypes, brain circuits and behaviours by simultaneously imaging and photostimulating hundreds of individual cells in 3D, with precise targeting of cells across large areas of the brain.
By selectively photostimulating only the target cells and rapidly switching between stimulation patterns, the user can understand neuronal interactions in networks. The product can also be used to specifically target cells and subcellular structures in layer 4 and beyond, achieving high-quality images. Users can quickly switch stimulation patterns to mimic neuronal firing, which should improve the accuracy of the data.
The high resolution of the imager allows cell bodies and subcellular structures to be specifically targeted with minimal off-target activation, providing clean data. Cells can be targeted in large brain networks to understand large neuronal networks.
The large field of view allows the use of a high magnification and resolution objective. Being able to excite or image target cells expressing different opsins or fluorescent proteins should increase experimental flexibility.
The modularity of the HyperScope and HoloStim-3D means the user can start with a Scientifica SliceScope upright microscope or Scientifica HyperScope multiphoton imaging system, and upgrade to SLM capability when required. The fully integrated hardware and software solution means users can start experimenting straight away.
