Protein synthesis is essential for cell growth, proliferation and survival, and can be an indicator of cellular stress and cell health. Unregulated protein synthesis may result in uncontrolled cell proliferation and growth, leading to diseases like cancer. As such, there is increasing interest in targeting protein synthesis as a part of cancer therapies.

The PURO-Fluor Protein Synthesis Labeling Kit from Cayman Chemical is a convenient, non-radioactive method for measuring protein synthesis by flow cytometry or fluorescence microscopy. The kit employs the cell-permeable, alkyne-containing puromycin analog O-Proargyl-puromycin (OPP).

Upon application to cells, the OPP probe incorporates into the C-terminus of translating polypeptide chains, thereby stopping translation. The truncated C-terminal alkyne-labelled proteins are subsequently detected via copper-catalysed click chemistry using 5 FAM-Azide.
