Cayman Chemical’s STAT3 TR-FRET kits use a homogeneous TR-FRET assay method for rapid measurement of phosphorylated and/or total STAT3 protein levels in cells.

STAT3 is a ubiquitously expressed transcription factor that has roles in a variety of cellular functions including proliferation, apoptosis and differentiation, as well as innate and adaptive immunity. Phosphorylation of cytosolic STAT3 at tyrosine 705 (Tyr705) in response to stimulation with cytokines or growth factors induces STAT3 dimerisation and nuclear translocation, leading to STAT3-dependent gene transcription.

Cayman offers three STAT3 TR-FRET kits: the STAT3 (Phospho-Tyr705) TR-FRET Kit measures the levels of STAT3 phosphorylated at Tyr705 (phospho-Tyr705); the STAT3 (Total) TR-FRET Kit measures total STAT3 levels; and the STAT3 (Total) and STAT3 (Phospho-Tyr705) TR-FRET Kit measures the levels of both. The streamlined ‘add-incubate-measure’ assay protocol is designed to decrease hands-on time and provide a powerful alternative to more error-prone and time-consuming techniques such as Western blot and ELISA.

The STAT3 TR-FRET kits exhibit low background fluorescence levels, a stable assay readout and high signal-to-background ratios. Each assay kit contains the appropriate TR-FRET antibody pair(s), as well as buffers and a positive control lysate. The robust assays are suitable for screening a large number of samples and are compatible with both 96- and 384-well formats.
