Oxford Expression Technologies pOET transfer plasmids

Oxford Expression Technologies has released four pOET transfer plasmids which encode for 6xHis-Tag fusion sequence and the pOET Sequencing Primers.

The vectors encode for combinations of either C- or N-terminal 6xHis-Tag fusion sequence and the insert either allows read-through in the correct reading frame or a fusion sequence that may be utilised if the insert includes a stop codon. Each vector has a Col E1 origin of replication and an ampicillin resistance gene for selection in E.  coli. The polh sequences have been replaced by a multiple cloning site (MCS) containing unique restriction sites for insertion of the foreign gene in the correct orientation. The range of pOET transfer vectors is designed for high-level expression of foreign genes under the powerful AcMNPV polyhedron (polh) promoter.

The pOET Sequencing Primers anneal to the upstream and downstream regions of the MCS and can be used to sequence across the MCS or amplify an insert within the MCS using PCR. The pOET range of transfer plasmids is compatible with any baculovirus system which utilises homologous recombination in insect cells, including the flashBAC range of products from Oxford Expression Technologies.
