ProteoStat Thermal shift stability assay kit
The ProteoStat Thermal shift stability assay kit includes a fluorescent dye which detects protein aggregation, so it can be used to monitor protein stability under systematic thermal stress conditions. From the thermal shift assay, a temperature at which the bulk of the protein becomes aggregated can readily be identified.
The aggregation temperature is an indicator of protein stability and can be used to optimise conditions that minimise protein aggregation as well as to identify ligands that bind and confer structural stability to a protein of interest.
Conditions that increase the aggregation temperature increase the stability of the protein.
The dye is a 488 nm excitable molecular rotor probe that, under standard aqueous conditions, is minimally fluorescent but, upon binding to the surface of aggregated proteins, emits a strong red signal at ~600 nm, providing a homogeneous assay for analysis of protein stability.
The assay facilitates analysis of protein stability over a wide range of pH values and ionic strengths, as well as using a variety of buffer, cryoprotectant and excipient compositions.
This facilitates understanding of the underlying mechanisms impacting protein stability, as well as predicting the stability of therapeutic proteins on long-term storage, screening the relative stability of protein variants (eg, amino acid substitution/deletion mutants, PEGylated proteins) and providing other critical information pertaining to the development of optimal protein formulations.
The assay can be performed using either a conventional fluorometer equipped with heating control or a real-time PCR instrument.
