In order to determine which novel compounds are warranted for further development, early detection of unwanted cytotoxic effects is vital.

Cytotoxic evaluation on cell lines has well established and widely accepted methods. In use are several different cellular targets using various read-outs and different exposure protocols. Toxicity testing at early stages of new product development should have high sensitivity, ie, effectively toxic substances should have a high probability of being identified. Tests should be reasonably simple, reproducible and cost effective.

The PAN I Cytotoxicity assay kit by Xenometrix allows applying four commonly used cytotoxicity assays sequentially on one cellular sample. It combines assays for extracellular lactate dehydrogenase (LDHe) to measure membrane integrity, the XTT tetrazolium salt to evaluate mitochondrial activity, neutral red (NR) for the evaluation of lysosomal activity and the sulforhodamine stain (SRB) for total protein content. This allows an investigator to identify the site of any cytotoxic damage.

This valuable information on cellular target structures can help decide which products are more suitable for further development and therefore where to focus investment resources.
