AGRF has incorporated two bioinformatics processing and analysis service options for its Contract Sequencing Service.
The most stringent assessment of sequence quality is an estimate of accuracy assigned to each base call in a trace. The program Phred is used to process trace data from AB377, AB3700, and MegaBACE sequencers. A Phred value of 20 or greater is considered to be high quality and means that a base call has a 99% or better chance of being accurate.
AGRF has developed software to process the raw Phred output into a graphical output file (in pdf format). This file shows the trace data and a colour-coded histogram of the corresponding phred quality scores for each sequence in a gel or plate run. In addition it provides a summary output of the quality for the entire gel/plate.
