Reversed-phase chromatography (RPC) does not work for everything. PolyLC has developed hydrophilic interaction chromatography (HILIC) columns that offer quick, isocratic separation of small, polar solutes normally eluting in the void in RPC, with online MS/MS quantitation.

This works for amino acids, polar metabolites etc. The columns can be used for the fractionation of peptides and hydrophobic proteins without detergents as well as the convenient elimination of detergents from microlitre-size peptide samples using inexpensive spin cartridges or coated pipet tips.

The ion-exchange columns are suitable for peptide separations, including the SCX-RPC sequence now used for proteome analysis. The unconventional use of ion-exchangers separates intact protein variants with 2-3x the resolution obtained with other columns. This eliminates the need for digestion and mapping.
