A gene expression technique adapted for single molecule sequencing has enabled researchers at the Riken Omics Science Center (OSC) to accurately and quantitatively measure gene expression levels using only 100 ng of total RNA. The technique, which pairs Riken’s Cap Analysis of Gene Expression (CAGE) protocol with the Helicos Genetic Analysis System opens the door to the detailed analysis of gene expression networks and rare cell populations.

Adenosine 3′, 5′ cyclic monophosphate (cAMP) plays a critical role in the transmission of signals by functioning as a ‘second messenger’. Binding of hormones or ligands to their respective receptors can either inhibit or enhance the production of cAMP by changing the activity of the enzyme adenylate cyclase. Adenylate cyclase is a membrane-associated enzyme that catalyses the formation of cAMP from ATP. Intracellular cAMP levels, in turn, regulate enzymatic activity of numerous protein kinases, which phosphorylate specific targets setting off a cascade of cellular events. Because of the importance of cAMP as a cellular messenger, several different reading methodologies have been developed to measure cellular levels, one of which is AlphaScreen. Here we describe and demonstrate the ability of the Synergy 2 to quantitate cAMP using this methodology.

Nucleic acids isolated from a variety of sample types such as animal models, body fluids or eukaryotic and bacterial cells provide the starting material for a broad range of downstream applications including gene expression, genotyping, sequencing and cloning. Quantification of isolated nucleic acid is necessary prior to introduction into these applications. The Epoch Spectrophotometer System provides a means of obtaining accurate measurement over a wide range of sample concentrations and volumes, including microvolume quantification. Comparison to other microvolume dedicated devices, such as NanoDrop, shows equivalent performance.
